How to use macros?

After some searching i can't find any help on applying macros, or even what they can and cannot do. The Wikipedia is no help.

How do i use them  :o :?:  
Any active links to get me started. I guess i'm more interested in using them with my VSTs but i also want to know what else they can do.

I agree that we should post this in the Wiki also.
