To make instruments sound more alive we'll need real-time pitch-shift of samples, without affecting their duration.
Also we'll need volume auto-compensation. Either automatic or by graph.
We'll need new instrument format.

We don't, really. That is what VST instruments are for. There are various VST samplers with real-time pitch shift or multi-layered instruments. For volume compensation, just put a compressor or whatever after the VST sampler.
I had talk about VST at Renoise forum.
They said they cant include VSTi into Renoise song file because of copyrights.
Not all VST are free, and those which are free not always free to redistribute, and those which are "double free" still will not be included into song file. LOL!!! :P

Sampler is not what we'll need.

And compressor is very bad idea. Because it compress bad notes but good notes too.

This is not possible without a more or less severe loss of sample quality, depending on the sample. It is technically simply not possible to do perfect pitch shifters and time stretchers.

There is Celemony Melodine sowtware, that seems to be able to do that.
Of course it is very complex. But only pitch algorithm is needed.

Melodyne is covered all kinds of patents and it isn't perfect, either.
